From 1980 to 1982, he competed in the American Wrestling Association (AWA). In the late 1980s, Santana and Canadian Rick Martel created the renowned tag team Strike Force.

He became the first Mexican American wrestler to win the WWF Intercontinental Heavyweight Championship on February 11, 1984. Santana competed in the first WrestleMania, defeating a masked wrestler known as The Executioner in the first bout at Madison Square Garden in March 1985. On August 8, 1993, he defeated longtime WWF opponent Don Muraco to win the ECW Heavyweight Championship. On March 13, 2004, he was inducted into the WWE Hall of Fame, with Shawn Michaels, his WrestleMania VIII opponent, giving the induction speech. In November 2010, Tito made an appearance in the Legends Roll-On.
